猪圆环病毒2型灭活疫苗中病毒抗原含量实时荧光定量PCR检测方法的建立

依据GenBank公布的猪圆环病毒2型Cap基因序列,在保守区域设计特异性引物和TaqMan探针,优化反应体系,建立评价猪圆环病毒2型灭活疫苗中病毒含量的实时荧光定量PCR检测方法,对方法的特异性、敏感性和重复性进行试验,并验证灭活剂用量和灭活时间对检测结果的影响。结果显示：该方法只对猪圆环病毒2型基因有特异性扩增,其他3种对照病毒基因的扩增结果均为阴性;检测灵敏度达到10^2.0 TCID₅₀/mL,比普通PCR方法高100倍;方法的重复性好,对同一样品进行10次检测,变异系数为2.28%;不同灭活剂用量和灭活时间对结果的影响较小,不会因各厂家使用的灭活剂用量和灭活时间不同,影响疫苗对比实验的公平性。该研究成功建立了一种评价猪圆环病毒2型灭活疫苗中病毒含量的实时荧光定量PCR检测方法,用于猪圆环病毒2型灭活疫苗样品中病毒抗原含量的定量,其结果可以反映不同猪圆环病毒2型灭活疫苗样品中抗原含量差异,为研究猪圆环病毒2型灭活疫苗病毒抗原含量评估方法提供了新的思路。     

2019—2020年新疆部分地区猪PCV2的抗体水平检测及分析

为了解2019—2020年新疆地区猪圆环病毒病2型（PCV2）抗体水平及其消长规律,本试验采用间接酶联免疫吸附试验（ELISA）方法对475份血清中PCV2免疫抗体水平进行检测和分析。结果显示,PCV2抗体平均阳性率为69.68%（331/475）,未达到国家规定标准（70%）。S/P的平均值为1.56。不同类别猪群抗体平均阳性率在42.50%~86.67%之间,有一定的差异。在调查的5个规模化养殖场中,仅有2个场PCV2免疫抗体阳性率达到国家标准。各场应根据抗体检测结果,进一步对现有的免疫程序进行调整和完整,确保各猪群健康。     

不同株型玉米苗期生长,光合速率及对肥料的反应

在人工气候室中，对盆栽条件下不同熟期的紧凑型与平展型玉米比较研究表明，单株玉米对土壤养分的反应株型间无规律性差异；不同熟期玉米间存在明显的规律性差异。植株苗期生长速率及高低土壤肥力下的差异与叶片Ｎ、Ｋ含量，光俣速率及不同光强，不同ＣＯ２浓度与的反应差异间存在明显的相关性。不同品种间苗期光合速率差异明显，土壤肥力高低对玉米苗期光合速率影响很大。     

Some Ideas on Reconstruction of Villages inside the City

In this paper, the historical reasons of formation of the village inside the city are analyzed. It is indicated that such situation for the villages inside the city are the result of joint action of both the villagers and the government. The foundation of villages inside the city laid on the simple compensation for farmers in confiscating the rural land and lack of intelligence aids. In this paper, the ideas and measures for their reconstruction are put forward based on consideration of the reasons of their formation and difficulties in the reconstruction.     

AN APPRONTMATE CALCULATING METHOD FOR WALL PLATES OF MAN'' S DRY GAS HOLDERS

The basic equations of wall plates of MAN' S dry gas holders are derived in this paper. According to the law of cylindrical bending given by the finite element results .we derive some approximate formula calculating the deflection and stresses of the wall plates from the basic equations. Compared with the known approximate calculating method.the method car. obviously increase the percision.     

Ecological Technology Strategy-the Typologic Analysis and Research of Double-Skin Facade

As a renovation envelope, double-skin facade has the characteristics of environmental function and unique aesthetic effect. In this paper, the composition and functional characteristics of double skin facade is introduced briefly, its development history and a detailed typologic analysis combined with typical cases is reviewed. It is pointed out that the double skin facade has a bright future in 21~(st) century and the function and aesthetics are the footstone for its typologic research.     

Structural Analysis and Discussion of Wide -stayed Bridge Basket

Based on a cable -stayed bridge girder as an example, the structure and function, the operating principle and calculation model of the main elements of the Basket of wide - stayed bridge were described in detail. A further discussion was carried out on the Basket of Cantilever construction of the main beam in the structure.
